Caffeine contracture in frog cardiac muscle after exposure to high concentrations of calcium.
نویسندگان
چکیده
Caffeine induced contracture in frog auricular muscle at room temperature when the muscle was exposed to a Ca-rich solution. The initial transient tension lasted 10-30 sec, and it was inhibited by procaine. It was dependent on caffeine concentration and the time of exposure to a Ca-rich solution prior to caffeine attack, but was little dependent on Ca concentration during caffeine contracture.

4-aminopyridine-induced contracture in frog ventricle is due to calcium released from intracellular stores.
The aim of the study is to demonstrate the presence of intracellular calcium store in frog ventricle based on contractures induced by 4-aminopyridine in calcium-free media. Frog-ventricular strips were subjected to field stimulation at 0.2 Hz and the force of contraction was recorded after stabilization. The preparation was then kept quiescent for some time in solutions with different sodium co...
متن کاملQuinine and Caffeine Effects on 45Ca Movements in Frog Sartorius Muscle
1 mM caffeine, which produces only twitch potentiation and not contracture in frog sartorius muscle, increases both the uptake and release of (45)Ca in this muscle by about 50 %, thus acting like higher, contracture-producing concentrations but less intensely. Quinine increases the rate of release of (45)Ca from frog sartorius but not from the Achilles tendon. The thresholds for the quinine eff...
متن کاملThe Relationship between Caffeine Contracture of Intact Muscle and the Effect of Caffeine on Reticulum
At concentrations between 1 to 10 mM, caffeine reduced the Ca-accumulating capacity of fragmented reticulum obtained from frog and rabbit muscle. With 8 mM caffeine enough Ca was released from frog reticulum to account for the force of the contracture. Caffeine did not affect all reticulum membranes equally. The fraction which was spun down at 2000 g was more sensitive than the lighter fraction...
متن کاملStudies of Cardiac Muscle with a High Permeability to Calcium Produced by Treatment with Ethylenediaminetetraacetic Acid
Thin strips of frog ventricle were isolated and bathed for 15 min in a solution containing 140 mM KCl, 5 mM Na(2)ATP, 3 mM EDTA, and 10 mM Tris buffer at pH 7.0. The muscle was then exposed to contracture solutions containing 140 mM KCl, 5 mM Na(2)ATP, 1 mM MgCl(2), 10 mM Tris, 3 mM EGTA, and CaCl(2) in amounts to produce concentrations of free calcium from 10(-4.8)M to 10(-9)M. The muscles dev...
متن کاملThe Time Course of the Loss and Recovery of Contracture Ability in Frog Striated Muscle Following Exposure to Ca-Free Solutions
Using area under the contracture curve to quantitate contractures, the diffusion coefficient of calcium ions within the frog toe muscle during washout in a calcium-free solution and subsequent recovery after reintroduction of calcium to the bathing solution was calculated to be about 2 x 10(-6) cm(2)/sec. The diffusion coefficient measured during washout was found to be independent of temperatu...
